Stasiun Tugu melayani jalur transportasi sejak 2 Mei 1887 yang awalnya hanya untuk transit dan bongkar muat hasil bumi. Sejak 1 Februari 1905, Stasiun Tugu baru digunakan untuk penumpang. Jalur luar kota pertama dibangun tahun 1899 menghubungkan Yogyakarta dan Surakarta. Stasiun Tugu adalah stasiun utama yang saat ini dikelola oleh PT Kereta Api Indonesia Daerah Operasi (Daop) IV Yogyakarta. Stasiun ini melayani perjalanan dengan kereta api kelas eksekutif dan bisnis jurusan Jakarta, Bandung, Purwokerto, Malang, Madiun, dan Surabaya. Kereta api komuter jurusan Kutoarjo, Solo, dan Madiun juga dilayani melalui stasiun ini.

Listed as national cultural heritage site in March 26, 2007 under the Decree of Minister of Education and Culture The Republic of Indonesia No. PM.25/PW.007/MKP/2007.
